How it works – The ProHisto Active Antibody Amplifier  System.

The four factors which most influence measurement of antigen-antibody  ( Ab-Ag)  reactions  in IHC and IF are :  Affinity ,Avidity , Relative Concentration and Antigen Solubility .  The vast majority of  primary antibodies are supplied in their monomeric form  and thus have ZERO native avidity . The Antibody Amplifier /Antibody system is designed to affect  primary antibody avidity and lattice formation thus making IHC and IF much more sensitive and reproducible than  previously possible

Avidity is promoted by the formation of polymeric antibody structures in physical forms such as rings or  links . These structures increase antibody binding  capacity well beyond the sum of individual monomeric antibody affinities  .  Prohisto’s technology creates an environment which may promote  avid structure formation among primary antibodies .

 

Concentration of primary antibodies  in the presence of target antigens affects the presence and structure of lattices composed of the avid antibody structures . Gross excess primary antibody concentration s such as 100:1 or 50:1 may prevent  or hinder lattice formation . ProHisto’s active antibody amplifier system creates a  relative [Ab/Ag]  concentration  greater than 1000:1 which may be conducive to lattice structure formation among avid structures .
