How it works – The ProHisto Active Antibody Amplifier System.
The four factors which most influence measurement of antigen-antibody ( Ab-Ag) reactions in IHC and IF are : Affinity ,Avidity , Relative Concentration and Antigen Solubility . The vast majority of primary antibodies are supplied in their monomeric form and thus have ZERO native avidity . The Antibody Amplifier /Antibody system is designed to affect primary antibody avidity and lattice formation thus making IHC and IF much more sensitive and reproducible than previously possible .
Avidity is promoted by the formation of polymeric antibody structures in physical forms such as rings or links . These structures increase antibody binding capacity well beyond the sum of individual monomeric antibody affinities . Prohisto’s technology creates an environment which may promote avid structure formation among primary antibodies . |
Concentration of primary antibodies in the presence of target antigens affects the presence and structure of lattices composed of the avid antibody structures . Gross excess primary antibody concentration s such as 100:1 or 50:1 may prevent or hinder lattice formation . ProHisto’s active antibody amplifier system creates a relative [Ab/Ag] concentration greater than 1000:1 which may be conducive to lattice structure formation among avid structures . |
